چکیده

In the fight against cancer, researchers have recruited an unlikely ally—a strain of Salmonella that causes food poisoning. Previously, scientists attempting to hijack this pathogen in order to target cancer struggled to balance its aggressive therapeutic actions and its harmful attacks on healthy body systems. Now, a team at the Helmholtz Center for Infection Research in Braunschweig, Germany proposed a more precise way to attenuate and strengthen this tumorfighting technique.

Live-Attenuated Bacterial Vectors: Tools for Vaccine and Therapeutic Agent Delivery

Genetically attenuated microorganisms, including pathogenic and commensal bacteria, can be engineered to carry and deliver heterologous antigens to elicit host immunity against both the vector as well as the pathogen from which the donor gene is derived. These live attenuated bacterial vectors have been given much attention due to their capacity to induce a broad range of immune responses inclu...

Engineered Salmonella enterica serovar Typhimurium overcomes limitations of anti-bacterial immunity in bacteria-mediated tumor therapy

Cancer is one of the leading causes of death in the industrialized world and represents a tremendous social and economic burden. As conventional therapies fail to provide a sustainable cure for most cancer patients, the emerging unique immune therapeutic approach of bacteria-mediated tumor therapy (BMTT) is marching towards a feasible solution. Although promising results have been obtained with...

Therapeutic vaccines based on genetically modified Salmonella: a novel strategy in cancer immunotherapy.

In the course of evolution, bacteria from the genus Salmonella adapted to survive and multiply in a vertebrate host. Skillful use of bacterial interactions with the host immune system became the basis for the development of modified Salmonella-based therapeutic vaccines. Bacterial genome can be modified to reduce toxicity and to develop or enhance therapeutic activity. Salmonella-based therapeu...

Bacteria - Fighters Against Cancer

200 years ago the observation was already described that tumors sometimes shrink when cancer patients undergo bacterial infections. This is the basis for bacteria‐mediated cancer therapy an alternative therapeutic approach of growing interest. Several bacterial strains are able to colonize solid tumors after systemic administration; a few even induce tumor reduction. Apparently, the virulence o...

Targeted Cancer Therapy Using Engineered Salmonella typhimurium

Obligate or facultative anaerobic bacteria such as Bifidobacterium, Clostridium, Salmonella, or Escherichia coli specifically colonize and proliferate inside tumor tissues and inhibit tumor growth. Among them, attenuated Salmonella typhimurium (S. typhimurium) has been widely studied in animal cancer models and Phase I clinical trials in human patients. S. typhimurium genes are easily manipulat...
